Dr. Jeffrey Gladden discusses Calocurb, a drug that appears to have lasting effects after you stop taking it. Patients don't experience rebound appetite or weight gain when they discontinue it. The drug seems to upregulate receptors that trigger appetite-suppressing hormones like GLP-1, CCK, and PYY, which stay elevated even after treatment ends.

It's really interesting that people come off of it and there isn't a rebound in appetite or weight gain. That's super exciting. And it also sounds like what I was hearing there, and you can correct me if I'm wrong, but that when you're upregulating, when you're increasing the stimulation of these receptors, you're also sort of upregulating their expression, as we talked about earlier, to the point where when you take it away, you're still making more GLP-1. CKK, PYY, than you would have been prior to taking it. And so you get sort of prolonged benefits even when it's not in your system. Is that true? That's what we're saying.
That's what we believe. I kind of like to think about it as that we've, and this may be too simplistic, but we've kind of exercised these little receptors. And so when you take the callicurb away, they're still upregulated.
