Scientist Stories: Jeff Litchman, Connectomics Across Scales

In this lecture Jeff Lichtman (Harvard) explores why the brain, unlike other vital organs, carries our sense of personhood by storing lifelong memories. He contrasts the common focus on synaptic plasticity with the stability of long‑term memory, illustrating the point with three YouTube demos: a motor‑memory bike‑riding task, the hollow‑face visual illusion, and the McGurk audio‑visual illusion. He then argues that learned behaviors become hard‑wired through experience‑dependent pruning of initially exuberant neural connections. Using his own early work on the rat submandibular ganglion, he shows how single axons start with many weak contacts that are later pruned to a few strong synapses. He extends this principle to cerebellar climbing fibers, motor‑neuron neuromuscular junctions, and cortical circuits, showing that early development features massive over‑connectivity that is sculpted into sparse, specific wiring. He describes modern connectomics tools—serial‑section EM, Brainbow mice, and AI‑driven segmentation (Google partnership)—that now allow reconstruction of thousands of neurons and millions of synapses, revealing non‑random clustering of multiple synapses from the same axon onto the same dendrite. Finally, he outlines the ambition of building a whole‑mouse connectome, the technical scale (exabyte data, multi‑beam microscopes, $100 M), and invites discussion on its scientific value. Throughout, he emphasizes that memory may be encoded not by adding connections but by eliminating the wrong ones, and that the brain’s wiring becomes simpler, not more complex, after birth.
