The NAD Mistake 99% of Biohackers Make (Molecular Biologist Explains)

In this Ben Greenfield podcast, molecular biologist Dr Nicola Conlon explains the biology of NAD⁺, why many biohacker supplements miss the mark, and how a systems‑based oral formulation can more effectively restore NAD⁺ levels. She outlines NAD⁺’s dual role as a redox co‑factor for mitochondrial ATP production and as a fuel (co‑factor substrate) for DNA‑repair enzymes, emphasizing the rule of thumb: high NAD⁺ = more energy + more repair. NAD⁺ is synthesized internally via the NAD⁺ salvage pathway, where the enzyme NAMPT recycles nicotinamide back to NAD⁺. With age, NAMPT activity declines roughly 50 % every 20 years, leading to a systemic NAD⁺ drop. Lifestyle stressors (exercise, fasting, caloric restriction) up‑regulate NAMPT, whereas chronic inflammation, sedentary behavior, poor sleep, and excess caloric intake suppress it. Inflammation also drives NAD⁺ consumption through the CD38 enzyme, which uses ~100 NAD⁺ molecules per catalytic cycle, far outpacing DNA‑repair enzymes. When the salvage pathway is impaired, nicotinamide accumulates and is shunted by the methyltransferase NNMT into methyl‑nicotinamide for excretion, creating a methyl‑donor burden. Simple precursor supplementation (NR, NMN) therefore plateaus and can increase methyl‑nicotinamide without raising NAD⁺, especially if CD38 is active. Dr Conlon critiques NAD⁺ IVs and patches, noting NAD⁺’s large, charged nature prevents cellular uptake, and high extracellular NAD⁺ can provoke inflammation. She then describes Nuchido TIME+, a proprietary oral blend that (1) supplies nicotinamide (the most bioavailable precursor), (2) directly activates NAMPT with rutin and indirectly via AMPK activation with alpha‑lipoic acid, (3) inhibits CD38 with apigenin (parsley extract), and (4) blocks NNMT‑mediated methylation with EGCG from green tea. A 2024 double‑blind, placebo‑controlled crossover trial showed that 7 days of TIME+ raised intracellular NAD⁺, increased NAMPT expression, reduced inflammatory cytokines, lowered glycated serum protein, reversed biological age by 1.26 years (GlycanAge), improved sleep, skin quality, energy, and sirtuin expression. The discussion also touches on dosing frequency (daily use is fine) and how the supplement complements hormetic stressors (heat, cold, sauna, fasting). The episode ends with promotional notes and a call for listener questions.
